Fourteen years later the same laboratory published a figure of 29.2 nmol per gram for whole rat brain. Both numbers describe the same molecule, cycloprolylglycine.

A tenfold disagreement inside one research programme is the first thing worth knowing here. The second is what the molecule is supposed to explain. Noopept is laboratory shorthand for N-phenylacetyl-L-prolylglycine ethyl ester, and the claim attached to it is a prodrug claim: the ester is cleaved in the body, and what actually reaches neural tissue is a smaller cyclic dipeptide. That proposed metabolite is cycloprolylglycine.

What the cycloprolylglycine metabolite hypothesis about noopept actually claims

PubChem lists the parent compound under identifier 180496, with molecular formula C17H22N2O4, a molecular weight of 318.4 and the systematic name ethyl 2-[[(2S)-1-(2-phenylacetyl)pyrrolidine-2-carbonyl]amino]acetate. That record carries a single registry number, 157115-85-0.

The proposed metabolite is smaller and far more polar: formula C7H10N2O2, molecular weight 154.17, a calculated partition coefficient of minus 0.6 and zero rotatable bonds. The identifier picture here is less tidy. PubChem holds the cyclic dipeptide under more than one record, one with unspecified stereochemistry and one specified as the 8aS configuration, and the stereospecified record carries two registry numbers rather than one. A single CAS number quoted for this molecule is a selection among records that PubChem itself does not merge.

The prodrug proposal separates into three claims that stand or fall independently: that cycloprolylglycine occurs naturally in brain tissue, that the parent ester is converted into it, and that the metabolite rather than the parent carries the pharmacology. The three rest on different evidence.

How cycloprolylglycine was first detected in rat brain

The 1996 report in FEBS Letters combined high-performance liquid chromatography (HPLC), gas chromatography and chromatography-mass spectrometry to identify the cyclic dipeptide in rat brain. The authors reported that synthetic material showed antiamnesic activity in a passive avoidance procedure, and proposed that its presence in brain pointed to a previously unrecognised route of memory regulation.

The metabolism paper followed in 1997 in the European Journal of Drug Metabolism and Pharmacokinetics. The parent ester was not detectable in rat brain one hour after intraperitoneal injection, down to the limit of detection of the HPLC method used. Three candidate breakdown products, phenylacetic acid, prolylglycine and cyclo-prolylglycine, were found in the brains of treated animals and also in untreated control animals. Only the cyclic dipeptide rose, roughly 2.5-fold, after the parent was given. Conversion in the presence of plasma and brain enzymes was also shown in vitro.

Two things follow from that experiment, and only two. The parent sat below detection in brain tissue, and one of its possible breakdown products increased. That pattern fits a prodrug relationship. It also fits rapid clearance of the parent, cyclisation occurring mainly in plasma, or a detection limit higher than the concentration present. The experiment did not separate those accounts.

Telling a parent ester apart from its cyclic breakdown product in a chromatogram needs retention time and mass together, which is why work of this kind runs against a characterised noopept analytical reference standard and synthetic cyclic dipeptide of declared stereochemistry.

Why the reported brain concentration figures do not agree

The 2010 localisation study from the same institute reported uneven distribution across brain regions in rats: 29.2 nmol per gram in whole brain, 38.9 in cortex and 126.4 in hippocampus. About 60 percent of the peptide appeared in the nuclear subcellular fraction.

Set that figure beside the 2.8 nmol per gram reported in 1996 and the discrepancy is an order of magnitude. Extraction chemistry, detectors and animal cohorts all move an absolute tissue concentration. The point is narrower: there is no settled endogenous concentration for this molecule, and no independent replication of either figure was located.

There is a further analytical wrinkle. Cyclic dipeptides of this class, the diketopiperazines, are not exclusive to mammalian tissue. A 2018 clinical study in Nutrients measured the same cyclic glycine-proline in cerebrospinal fluid and reported that the molecule was already present in the blackcurrant preparation given to participants. A compound occurring in plant material has a possible exogenous contribution to tissue concentration, which complicates the word endogenous rather than refuting it.

What the AMPA receptor and TrkB experiments measured

The mechanistic case built slowly and in one place. A 2016 paper in Doklady Biochemistry and Biophysics reported that the cyclopeptide at one micromolar enhanced transmembrane AMPA currents in rat cerebellar Purkinje cells, and described it as an endogenous positive modulator of AMPA receptors, linking that to earlier work on brain-derived neurotrophic factor in culture.

Two blockade studies followed. In 2020 the same group reported that DNQX, an AMPA receptor antagonist, and K252a, a Trk receptor inhibitor, each prevented the anxiolytic effect in rodents. In 2022 they reported that DNQX, the non-competitive blocker GYKI 52466 and K252a each prevented the neuroprotective effect in vitro. Blockade experiments speak to necessity and are silent about sufficiency: they show an intact AMPA and Trk path is required, not that the peptide binds either receptor. No binding constant is reported anywhere in the series.

A separate 2020 study looked for mitogenic activity in HEK293 and SH-SY5Y cells and found none, reporting a reduction in early apoptotic cells instead. That negative result was published openly.

One 2022 finding does not sit comfortably inside a clean prodrug account. A group at the Research Center of Neurology in Moscow, working in rat hippocampal slices, reported that the parent dipeptide itself increased firing of GABAergic interneurons, and that alpha-bungarotoxin and methyllycaconitine, both selective for the alpha7 nicotinic acetylcholine receptor, almost completely abolished the effect. In an acute slice the parent molecule is doing something of its own.

The same molecule arrives from a completely different direction

Outside the Russian literature the identical cyclic dipeptide is studied under a different name and a different origin story. Work from the University of Auckland treats cyclic glycine-proline as a metabolite of insulin-like growth factor 1, not of any synthetic ester.

A 2014 paper in Scientific Reports reported that the cyclic peptide alters binding of IGF-1 to its binding protein IGFBP-3, promoting IGF-1 activity when the growth factor is scarce and damping it when abundant. A 2019 study measured the peptide in plasma from 34 stroke patients and 50 age-matched controls, finding the molar ratio to IGF-1 lower in patients at baseline and rising over 90 days. The 2018 cerebrospinal fluid study involved 11 participants.

These are small datasets, described by their authors as exploratory biomarker work rather than as mechanism. Their relevance here is indirect but real: they give the molecule an endogenous route, via cleavage of a growth factor, with no synthetic compound in it.

Line of evidence

What was measured

What it does not settle

1996 identification

Presence in rat brain, three chromatographic methods

Absolute concentration; independent replication

1997 metabolism

Parent below detection, cyclic product raised 2.5-fold

Where cyclisation occurs; whether the parent is inactive

2016 electrophysiology

Enhanced AMPA currents at one micromolar

Binding site, affinity, selectivity

2020 and 2022 blockade

Antagonists abolish two measured effects

Sufficiency; order of events in the pathway

Where the evidence stops short of an established mechanism

Concentration of authorship is the largest structural weakness. Almost every primary paper naming the cyclopeptide as a mnemotropic agent carries an affiliation to the Zakusov Research Institute of Pharmacology in Moscow, and most appear in two translated short-format journals. That is a coherent programme rather than a distributed field.

The metabolite claim also generalises in a way that weakens its specificity. A 2022 paper on a different proline-containing ester, GZK-111, reported four biotransformation products and again named the same cyclic dipeptide among them. A metabolite common to a chemical series is a plausible product of that series, not evidence that it is the active species for any one member.

The gap that matters most is human. No pharmacokinetic study showing that the cyclopeptide rises in human plasma or cerebrospinal fluid after the parent ester was located, and the Auckland measurements concern the peptide arriving from growth factor cleavage instead. Read strictly, the literature supports a narrow statement: the cyclic dipeptide is detectable in rodent brain, it increases after the parent ester enters the animal, and its effects in two rodent assays require intact AMPA and Trk signalling. Every step past that is a hypothesis with an experiment still attached.

Frequently asked questions

What is the cycloprolylglycine metabolite of noopept?

It is a cyclic dipeptide, formula C7H10N2O2 and molecular weight 154.17, formed by cyclisation after the parent ester bond is cleaved. A 1997 rat study found the parent below detection in brain while this product rose about 2.5-fold, which is the origin of the prodrug proposal. The conversion remains undemonstrated in humans.

Is cycloprolylglycine an endogenous brain peptide?

It was reported in untreated rat brain in 1996 and again in a 2010 regional study, so it is present in tissue that received nothing. Two complications remain: the two reported whole-brain concentrations differ roughly tenfold, and the same cyclic dipeptide occurs in plant material, so a dietary contribution to tissue levels cannot be excluded.

How much cycloprolylglycine is in rat brain tissue?

Published figures disagree. The 1996 identification paper reported 2.8 nmol per gram of wet brain by gas chromatography. The 2010 localisation study from the same institute reported 29.2 nmol per gram in whole brain, 38.9 in cortex and 126.4 in hippocampus, with about 60 percent of the peptide in the nuclear subcellular fraction. No independent replication was located.

Does cycloprolylglycine modulate AMPA receptors?

One electrophysiology study reports that it does. In 2016 a Moscow group measured enhanced transmembrane AMPA currents in rat cerebellar Purkinje cells at a concentration of one micromolar and described the peptide as a positive endogenous modulator. No binding affinity has been published, and the result has not been reproduced by an independent laboratory.

Is cyclic glycine proline the same molecule as cycloprolylglycine?

Yes, the two names describe the same cyclic dipeptide. A New Zealand research group studies it as a metabolite of insulin-like growth factor 1 and as a plasma biomarker, while a Russian group studies it as a nootropic metabolite. The two literatures use different nomenclature, different assays and largely separate citation networks.

Has the noopept prodrug hypothesis been replicated independently?

Not in the strict sense. Almost all primary work naming this metabolite as the active species originates from one Moscow institute and appears in two translated journals. An independent 2022 slice study found the parent dipeptide itself acting through alpha7 nicotinic receptors, which fits awkwardly with the idea that it is only a carrier.

References

Gudasheva et al., FEBS Letters, 1996 – cyclo-prolylglycine in rat brain

Gudasheva et al., Eur J Drug Metab Pharmacokinet, 1997 – major metabolite of GVS-111

Boiko et al., Bull Exp Biol Med, 2010 – localization in rat brain

Gudasheva et al., Dokl Biochem Biophys, 2016 – positive modulator of AMPA receptors

Gudasheva et al., Dokl Biochem Biophys, 2020 – anxiolytic effect, AMPA and TrkB

Gudasheva et al., Dokl Biochem Biophys, 2022 – neuroprotection, AMPA and TrkB

Zainullina et al., Bull Exp Biol Med, 2020 – cell proliferative activity

Kondratenko et al., Neuroscience Letters, 2022 – alpha7 nicotinic receptors in CA1

Kolyvanov et al., Bull Exp Biol Med, 2022 – metabolism of GZK-111 in rats

Guan et al., Scientific Reports, 2014 – cyclic glycine-proline regulates IGF-1 homeostasis

Fan et al., Ann Clin Transl Neurol, 2019 – plasma ratio in stroke patients

Fan et al., Nutrients, 2018 – cyclic glycine-proline in cerebrospinal fluid

Research use only. Nonsensia supplies analytical reference standards for laboratory and research applications.
